Omeprazole is a highly effective, well-tolerated, and economically feasible PPI for acid peptic disorders, which improves GERD symptoms and heals erosions and ulcers quickly.

Omeprazole continues to stand out as a leading proton-pump inhibitor (PPI) for treating acid peptic disorders (APDs) such as gastroesophageal reflux disease (GERD) and ulcers, demonstrating both efficacy and tolerance. Notably, it also emerges as the most cost-effective option, as per described by a systematic review and meta-analysis published recently in ‘The International Journal of Clinical Practice’.

This comprehensive analysis aimed to compare omeprazole’s efficacy against other PPIs and placebos in treating APDs. Databases such as Google Scholar, PubMed and China National Knowledge Infrastructure were searched up to March 2023 for related clinical trials. The analysis focused on both erosive and non-erosive diseases, with primary outcomes including symptom improvement and ulcer or erosion healing rates. Adverse events, cost-effectiveness, nighttime acid build-up and hospitalization were the secondary outcomes.

Thirty-one trials with a total of 10,539 individuals (including 12 studies) were counted. Omeprazole significantly outperformed placebo in reducing heartburn (RR=2.47, 95% CI: 2.13-2.86, p<0.001) and was on par with other PPIs. It also had a lower incidence of adverse events compared to both placebo and other PPIs (11% versus 31%, respectively). Moreover, omeprazole proved to be the most cost-effective option, particularly in the Indian market.
